Road Incident Management System (RIMS) chairperson Sipho Towa says the road will remain closed for several hours while recovery and rehabilitation work is carried out.
Motorists have been urged to avoid the area and not attempt to pass the crash scene.
Authorities have advised motorists travelling from Bloemfontein to use the N1 south and exit at Springfontein or Bethulie before rejoining the N6.
Drivers travelling from Smithfield are advised to take the R701 via Bethulie to Springfontein and then join the N1 north towards Bloemfontein.
Traffic officials say the closure will remain in place until the heavy vehicle has been recovered and the roadway has been cleared and declared safe for use.
